However, there’s evidence that this increase in NAD+ production tapers off after 2-3 months of supplementation. In one NMN study, NAD+ levels peaked at week eight but tapered off by week 12. And an NR study in 140 participants saw NAD+ levels peak at 14 days and drop slightly by day 56 in all three dose groups (100mg, 300mg, and 1,000mg).

The likeliest culprit of NAD+ precursors’ time-limited efficacy lies in the metabolic process they affect most directly: the salvage pathway. We’ll go way deeper into the mechanics of the salvage pathway later in this guide, but the fundamental issue with NAD+ precursors is that, by increasing NAD+ levels, they also increase the byproducts of NAD+ utilization, which recycle back into the salvage pathway and demand more fuel to turn into NAD+ once again.

Without adding the appropriate fuel alongside a given NAD+ precursor, the works eventually gum themselves up. Innerbody Labs puts this fuel back into the process by providing a 500mg dose of trimethylglycine (TMG) in every serving. Also known as betaine, TMG acts as a critical methyl donor to NAD+ production, ensuring the salvage pathway continues to operate at an increased pace. The added 15mg of spermidine plays a similar role, staving off an enzyme that could hijack NAD+ byproducts out of the salvage pathway and convert them into unhealthy metabolites.

Rounding out the formula, 10mg of BioPerine is twice the dose that was shown in one impressive study to increase NMN absorption and conversion to NAD+ by around 15%. Innerbody Labs NAD+ Support gets high marks for safety for a number of reasons. For one, as we discussed above, the inclusion of spermidine in the formula helps prevent NAD+ byproducts from being misappropriated. Specifically, we’re referring to spermidine’s ability to suppress nicotinamide N-methyltransferase (NNMT), which can turn nicotinamide (NAM, a byproduct of NAD+ use) into 1-methylnicotinamide, a substance associated with fat accumulation and insulin resistance.

NAD+ Support also introduces a little extra safety by splitting its precursor dose between NMN and NR and by keeping that combined dose within safe limits. Research has shown that NR doses up to 2,000mg/day and NMN doses up to 1,200mg/day are safe and yield only occasional GI side effects. NAD+ Support’s 400mg of NR and 700mg of NMN together equal 1,100mg total — more than most competitors’ precursor totals without reaching into unsafe territory.

NAD+ is a coenzyme that helps fuel and manage metabolic activity in the mitochondria — the portion of a cell that turns the food you eat into energy. How Innerbody Labs NAD+ Support works

NAD+ Support relies on the two most well-regarded and widely studied NAD+ precursors available: NMN and NR. It also includes a pair of ingredients with impressive track records in longevity research: TMG and spermidine.

Here’s how those four ingredients — and a fifth, BioPerine — have performed in NAD+ and longevity research:

NMN

NMN has been shown to increase NAD+ levels at doses of 300mg/day, with a significant jump at 600mg/day and a modest increase above that at 900mg/day. That implies that somewhere between 600mg and 900mg is the most efficacious dose for the average person to increase their NAD+ levels. Other doses have shown impressive secondary effects, including a 250mg dose to improve sleep and a 300mg dose to increase telomere length (protective caps on DNA, the lengths of which correlate to aging).

NR

A 2019 study of NR’s effects on NAD+ levels showed a positive impact starting at just 100mg, which increased NAD+ levels by 21%. That same study showed that a 300mg dose increased levels by over 50% and a 1,000mg dose increased levels by over 140%. Secondary effect research has produced improvements in biomarkers for neurodegenerative disease and blood pressure, both at 1,000mg.

TMG

TMG is included in this formula as a methyl donor to fuel the salvage pathway, ensuring sustained NAD+ production beyond the drop-off seen in both NR and NMN research. Beyond balancing against NNMT, spermidine has been linked in massive cohort studies with increased longevity, and in smaller studies with improved autophagy, bolstering cellular health overall.

BioPerine

BioPerine doesn’t boast abilities to boost NAD+ or bolster longevity, but it’s well-known for its ability to improve nutrient absorption. For NMN, specifically, 5mg of BioPerine has been shown to increase NAD+ production by just over 15%.

Taken together, these five ingredients have the potential to increase NAD+ production, provide secondary benefits associated with both NMN and NR, deliver longevity enhancements from spermidine and TMG, and prevent a drop-off in NAD+ production often seen in long-term precursor supplementation.

Some competitors include similar ingredients in their formulas, like AEON from Perpetua.Life. AEON also combines NR with NMN and includes TMG, spermidine, and black pepper fruit extract. However, in all but the last of those ingredients, AEON’s doses are less than half of what Innerbody Labs offers.

A crash course in NAD+ production

Understanding how the ingredients in NAD+ Support come together to increase NAD+ levels sustainably requires some understanding of NAD+ metabolism.

NAD+ precursors almost all rely on one metabolic system for NAD+ production: the salvage pathway. The other two pathways for producing NAD+ are the Preiss-Handler and de novo synthesis pathways, which rely on dietary vitamin B3 and tryptophan, respectively, and only account for about 15% of all NAD+ production. That means that 85% of all NAD+ production occurs via the salvage pathway.

The Role Of TMG And Spermidine In The Salvage Pathway

In the salvage pathway, NAM combines with 5-phosphoribosyl-1-pyrophosphate before becoming NMN, then turning into NAD+. When you take NMN, it jumps ahead of NAM and turns into NAD+ directly. When you take NR, it turns into NMN before becoming NAD+.

You’ll notice TMG and spermidine in the diagram above, as well. TMG’s role is in fueling NAM’s conversion to NMN. Without enough methyl donors like TMG in the picture, increases in NAD+ production can fade after around 12 weeks.

Meanwhile, spermidine fends off NNMT, which is trying to divert a bit of NAM from the salvage pathway and repurpose it for what ultimately might do more harm than good.

With these four ingredients in supplement form (NMN, NR, TMG, and spermidine), NAD+ production can increase and sustain itself.

Why combine NMN and NR?

As we alluded to above, NMN and NR enter the salvage pathway at different points. They also rely on different processes and transporters to get into cells before becoming NAD+. By supplementing with both, NAD+ production efficiency should be greater than if you were to supplement with just one or the other.

The only problem with this hypothesis is that it hasn’t been tested directly. There aren’t any studies pitting combined precursor administration against a monotherapy of one or the other.

Despite the data that’s missing from the research landscape, there are compelling reasons to combine precursors, and they come from the studies of each individual ingredient: NMN research has revealed certain secondary effects that NR studies haven’t produced, and vice versa.

For example, NMN’s abilities to improve sleep and telomere length haven’t been reproduced in NR studies. And NR’s ability to improve biomarkers of neurodegenerative disease hasn’t been reproduced in NMN research. It’s entirely possible that these effects are linked more to increased NAD+ levels than to either precursor, but if you look at where the science is today, taking both should offer a greater range of potential benefits than taking just one.

Who is NAD+ Support for?

Innerbody Labs NAD+ Support is ideal for anyone concerned with aging and age-related NAD+ decline. At this point, exact rates of decline are still being studied, making it hard to determine when you really need to worry about your NAD+ levels. Several papers have connected NAD+ decline with middle age, with 45 being the point when significant declines become evident.

That point in time tracks with a more recent paper illustrating two distinct periods in life when aging takes a jump in most people: at around 44 and around 60 years of age. So, NAD+ Support is probably best suited to anyone 40 and above, though it may also provide protective benefits to younger people concerned with their eventual decline.

Who might want to look elsewhere?

As we just discussed, NAD+ levels are likely rather high in most young adults, making an NAD+ booster less of an ideal supplement for them. There are also some areas where NAD+ boosters have shown promise but lack a wealth of research behind them.

For example, while NR has shown potential to improve certain biomarkers of neurodegenerative disease, it isn’t a prime candidate for treating dementia. Similarly, although NMN has the potential to improve sleep, it isn’t a better choice than a good OTC sleep aid.

At best, NAD+ boosters could serve as supportive aspects of a multipronged approach to tackling these issues in consultation with your doctor. For now, boosting NAD+ levels remains the primary target of NMN, NR, and Innerbody Labs NAD+ Support.

Is Innerbody Labs NAD+ Support safe?

Innerbody Labs clearly considered the available research when formulating NAD+ Support, as its combined precursor dose comes in at just under the 1,200mg level that’s been shown safe for NMN consumption. The same can be said when comparing it to NR doses reaching above 2,000mg with safe results. Even with the 15% boost in NMN efficacy provided by BioPerine, and assuming that a similar boost applies to NR, you only get to an equivalent precursor dose of 1,265mg.

The other ingredients all have good safety profiles, including:

All that said, there are some unanswered questions surrounding any NAD+ precursor’s long-term safety, especially as it relates to cancer risk, with research seeming to point in opposite directions. Some research has pointed toward NAD+ restrictions as a potential therapy to starve tumor growth, implying that excess NAD+ could do the opposite, fueling tumor growth in already present cancer. However, other research has shown that increased NAD+ levels inhibit tumorigenesis in the first place.

It’s possible that both are true: that NAD+ can inhibit initial tumor growth and also increase tumor intensity if levels rise after a tumor has formed. Much more research is needed in this field, and it makes it all the more important for you to talk with your doctor before adding any NAD+ precursor to your regimen.

Exercise

Along with diet, exercise has also been shown to improve NAD+ levels. Specifically, a study in untrained individuals found that resistance training improved NAD+ levels in the muscle and throughout the body.

Sleep
